Treating a certain type of prostate cancer that has spread to other parts of the body. It is used along with another medicine (prednisone). It may also be used for other conditions as determined by your doctor.
Xbira (Abiraterone Acetate ) is an androgen biosynthesis inhibitor. It works by decreasing levels of certain hormones produced by the testes. This helps to prevent the growth of certain tumors that need these hormones to grow.
Mechanism of action:
Xbira (Abiraterone Acetate), the active metabolite of Xbira (Abiraterone Acetate ), inhibits CYP17A1, which manifests as two enzymes, 17-hydroxylase (IC50 = 2.5 nM) and 17,20-lyase (IC50 = 15 nM) (six-fold more selective for inhibition of 17-hydroxylase over 17,20-lyase) that are expressed in testicular, adrenal, and prostatic tumor tissues. CYP17 catalyzes two sequential reactions: (a) the conversion of pregnenolone and progesterone to their 17-hydroxy derivatives by its 17-hydroxylase activity, and (b) the subsequent formation ofdehydroepiandrosterone (DHEA) and androstenedione, respectively, by its 17,20-lyase activity. DHEA and androstenedione are androgens and precursors of testosterone. Inhibition of CYP17 activity by abiraterone thus decreases circulating levels of androgens such as DHEA, testosterone, and dihydrotestosterone (DHT).
Abiraterone also acts as a partial antagonist of androgen receptors, and as an inhibitor of the enzymes 3-hydroxysteroid dehydrogenase, CYP11B1 (steroid 11-hydroxylase), and other CYP450s (e.g., CYP1A2, CYP2C9, and CYP3A4).
Xbira (Abiraterone Acetate ) reduces serum testosterone levels to less than 1 ng/dL (i.e., undetectable), and decreases the weights of the prostate gland, seminal vesicles, and testes, in accordance with its antiandrogen action.
Dosage and Administration:
*Xbira (Abiraterone Acetate ) is a tablet, taken by mouth. It is taken once a day.
*It must be taken on an empty stomach [2 hours after a meal, or one hour before]
*It should be taken at approximately the same time each day.
*Prednisone [10mg daily] should be taken along with Xbira (Abiraterone Acetate ).
*The amount of Xbira (Abiraterone Acetate ) you receive depends on many factors. Your doctor will determine your dose and schedule
All medicines may cause side effects, but many people have no, or minor, side effects. Check with your doctor if any of these most COMMON side effects persist or become bothersome:
Cough; diarrhea; heartburn; hot flushes; joint swelling or pain; mild muscle discomfort; tiredness; vomiting; weakness.
Before starting Xbira (Abiraterone Acetate ) treatment, make sure you tell your doctor about any other medications you are taking (including prescription, over-the-counter, vitamins, herbal remedies, etc.). Do not take aspirin, products containing aspirin unless your doctor specifically permits this.
Xbira (Abiraterone Acetate ) is currently not indicated for use in women. However, if Xbira (Abiraterone Acetate ) is given to a woman, getting pregnant should be avoided and the woman should not breast feed.
Store: Store Xbira (Abiraterone Acetate ) at room temperature, between 59 and 86 degrees F (15 and 30 degrees C). Store away from heat, moisture, and light. Do not store in the bathroom. Keep Xbira (Abiraterone Acetate ) out of the reach of children and away from pets.
